Abstract: An experiment was conducted to investigate the effect of genotype meat-type or egg-type ; on the AME digestibility of selected ingredients. The practice of using only one value for apparent metabolizable energy AME ; of feed ingredients for all classes of poultry has been questioned. The ability to convert nutrients to the final products depends on a variety of factors, such as age, sex, genetic and housing system. Some studies have indicated that broiler breeders are less able to metabolize diet energy, than are Leghorn birds. In this study, the ingredients included samples of cereals corn, wheat and barley ; , one sample of plant protein soybean meal ; and one cereal by-product wheat bran ; . Forty adult males 20 per breed ; were randomly assigned to five feedstuffs. Each feedstuff was fed to four birds per breed. The AMEn values were determined using a classical total collection method Sibbald method ; . The AME of each feed ingredient was calculated from the difference between GE intake and GE losses in the droppings. The AME values were corrected to zero nitrogen balance AMEn ; . The results were showed the average AMEn digestibility in wheat egg-type : 2785, Kcals Kg ; meat-type : 2750 Kcals Kg ; and barley egg-type : 2715 Kcals Kg; meat-type : 2675 Kcals Kg ; samples were similar in two breeds. The AMEn digestibility in White Leghorn birds was significantly greater for corn egg-type : 3065 kcals Kg ; meat-type : 2842 kcals Kg ; , soybean egg-type : 2185 Kcals Kg ; meat-type : 2040 Kcals Kg ; and wheat bran egg-type : 1440 Kcals Kg ; meat-type : 1333 Kcals Kg ; compared to the broiler strain. In this study, considerable differences were also noted between the two breeds that were tested in their ability to utilize the AME of feed ingredients. In mature broiler breeder hens excess ME intake can cause fat deposition; consequently, accurate representation of the energy content of a breeder diet is important. On the other hand, deficiencies of energy around the time of peak egg production will likely reduce egg production at this time, or production will fall after peak by definition. Since tables concerning ingredient composition usually contain ME values determined with White Leghorn, further research is needed if energy content of broiler breeder diets is accurately represented. Abstract 214088: "A safety and efficacy study of a novel formulation of prednisolone metasulfobenzoate PREDOCOL ; in the induction of remission and maintenance in ulcerative colitis" This is a safety and efficacy study of a novel formulation of prednisolone metasulfobenzoate PREDOCOL ; in the induction of remission and maintenance in ulcerative colitis. The background here is that there is an almost 20-year-old study showing that prednisolone metasulfobenzoate is topically effective and really very poorly absorbed from the colon. The idea here was to take this poorly absorbed agent, encapsulate it in an Eudragit L capsule which releases the drug in the ileum or thereabout, put about 200 controlled release drug pellets into this capsule and see if this would be beneficial. The idea of treating with steroids is we give it as a taper. We treat, we taper down, and the idea is to get the patient off of the agent. Here the notion is longer term treatment with a poorly absorbed steroid, essentially. In this study, 59 patients got PREDOCOL 40 mg per day for six months, another 61 patients were assigned to PREDOCOL 60 mg a day ; for six months, and then the third arm was actually an active arm, 61 patients assigned to prednisolone 40 mg a day ; with a tapering regimen over eight weeks. All this was given as a BID divided dose. They looked at steroid side effects by visual analog scale. They also measured the Powell-Tuck score, which was one of the ulcerative colitis indices, physician global assessment, and sigmoidoscopy by Baron score. The design was intended to be non-inferiority in terms of efficacy to prednisolone taper. The long and short of it is that at least for the 40 mg group, it appeared that there was a remission benefit over the six months of treatment. I found this to be really intriguing, similar to what has recently been done with asthma; give longer term steroids to suppress the disease. We don't do that in inflammatory bowel disease because of the long-term side effects. If we could give steroids continuously and in a safe fashion, would we want to do that? Would that actually make patients feel well? Would it heal the mucosa and keep it healed? I don't know but this sort of opens that question because it looks like this might be relatively safe. There is certainly a lot more that we have to learn about this. We need to learn about suppression of the hypothalamic pituitary adrenal axis and lots of other things. Canadian sites participating in FDG-PET in studies must enroll patients in the Health Canada Safety Awareness Protocol or another Health Canada approved clinical study for FDG-PET and provide a copy of the institution's "No Objection Letter" for that study to RTOG with study regulatory documents. 6.11.1 PET Image and Scanner Compatibility Requirements All imaging must be conducted on a combined PET CT scanner with full ring PET and four slice or greater multi-detector CT. The scanner should be operating in high-sensitivity 2D mode, if available. 3D mode is permissible for patients imaged on combined PET CT scanners without a 2D mode. Note: Scanners with less than four slice capability may be acceptable, but these scanners must be reviewed by the PET Co-Chair, David Schwartz, MD, on a case-by-case basis. Patients will observe a four to sixhour fasting period prior to FDG injection. Patients with a history of medically controlled diabetes will be counseled to check serial blood sugars prior to each scan to ensure that values average below 200 mg dL. For these patients, a blood sugar measurement will be performed after a six-hour fast to gauge fasting tolerance. Serum glucose concentration will be measured for all patients prior to scanning and must be less than 200 mg dL to proceed to imaging. FDG Injection A dose of 10-20 mCi of 18FDG will be infused intravenously. As per best clinical practice, administration of 0.5 mg of alprazolam 5-15 minutes prior to FDG injection to relax the patient and to reduce neck 18FDG muscle uptake should be considered. The patient will lie quietly for at least 30 minutes, and scanning will begin 50-70 minutes following the FDG injection. PET Imaging Imaging must encompass the vertex of the head down through the entire pelvis. The recommended imaging protocol incorporates two discrete phases, and is as follows: During the first phase, head and neck scanning will be performed with full neck extension. The patient will initially be imaged with a 120 KeV 300 mA, 0.5-second detector rotation time "high mA" ; CT scan with intravenous contrast 100 cc contrast bolus administered at 1.5 cc second, with a 50 second scan delay and with the scan started inferiorly, moving cranially ; , followed by a 120 KeV 80 mA, 0.8-second detector rotation time "low mA" ; CT scan for PET attenuation correction, followed lastly by PET scanning. Alternatively, an initial low mA CT scan may be performed for attenuation correction, followed by a high mA CT scan with intravenous contrast. Standard manufacturer recommendations for specific low and high mA CT scanning parameters can be substituted for those listed above. Two fields of view approximately 15 cm ; will be used for head and neck PET imaging. Patients then will be allowed to rest their necks for 1-2 minutes. For the second phase of imaging, the neck will be shifted into neutral position, and the remainder of the body will be surveyed per routine local institutional protocol with arms raised above the head to allow for optimal thoracic and upper abdominal imaging.
