The in vitro effects of the following antimicrobial agents on Toxoplasma gondii tachyzoites were studied: artemisinin ether arteether ; , cycloguanil hydrochloride cycloguanil ; , mefloquine, primaquine phosphate, and quinine sulfate, as well as the calcium channel blocker verapamil and the calmodulin inhibitor trifluoperazine hydrochloride. Arteether at .0.5 , ug ml and cycloguanil at .1.0 , ug ml inhibited T. gondii in vitro. Cycloguanil 2.5 , ug ml ; combined with a noninhibitory concentration of sulfadiazine 25 , ug ml ; inhibited T. gondii more than cycloguanil alone. Neither primaquine phosphate, mefloquine, nor quinine sulfate had an inhibitory effect on intracellular T. gondii. Verapamil and trifluoperazine hydrochloride were not inhibitory at lower physiologic concentrations, but higher physiologic concentrations were toxic to cell cultures in vitro and therefore our assay could not be used to assess their effects.

He also presented the work of the group in Marseille, France on muscle energetics in MHS patients, some of whom had history of exertional heat stroke EHS ; . Compared to MHN subjects there were a greater decrease in pH and PCr during exercise in MHS + - EHS. The was no difference in post-exercise recovery between groups. Furthermore, the Marseille group reported on 376 subjects with exertional heat stroke who were evaluated with IVCT, histochemistry and 31PNMR. These episodes of EHS occurred during military exercises. There was no clear relationship to environmental temperatures, which ranged from 0C to 32C in 60% of EHS episodes. This resolved without treatment. Two days later, he reported nausea, vomiting and back pain. This was treated with diclofenac. These were the only factors potentially related to muscular and renal injury at the time he presented seven days post-operatively with creatinine of 8.8 mg%. No abnormalities had been noted in the first three days post-operatively. He was found to have MH susceptibility by the European protocol for muscle contracture testing seven months later. Stefan Mohr of Ullevl Univ., Tjome, Norway, described the presentation of MH during cardiopulmonary bypass of a 56-year-old man with a history of smoking, heavy alcohol intake and hypertension whose only medication was an ACE inhibitor presented for aorto-coronary grafts. After initiation of bypass, anesthesia was initially continued with sevoflurane through the bypass machine. Epinephrine infusion was needed to maintain blood pressure. Nevertheless, flow required to maintain perfusion was 75% above expected. Blood gas demonstrated mixed acidosis. Gas flow through the bypass reservoir was increased by 50% above the expected value to maintain acceptable pCO2. Lactic acid was elevated. Peak temperature was 38C. Peak CK was 38, 000 iu. The family requested muscle biopsy. Four weeks after surgery, the IVCT was performed showing contracture of 3 grams with 2% halothane and 0.3 grams with 2 mM caffeine. Laboratory Studies: There were a number of studies of contracture testing methods, including that of Lucas Weigl and Hans Kress in Vienna, Austria, who did not find 4-chloroM-cresol 4CmC ; to improve discrimination between MHS and MHN muscle cells. Martin Anetseder presented more doseresponse studies demonstrating differentiation of MHS from MHN in 3 hind limb muscles of pigs. Caffeine had a greater effect than halothane, but both produced greater increases in lactate in MHS animals.
